Selection and Verification of Reference Genes for Gene Expression Studies in Different Cell Lines of Golden Pompano (Trachinotus ovatus)

The golden pompano snout (GPS) and head kidney (GPHK) cell lines have been proven to be meaningful tools for the study on pathogenic infections in vitro. In this study, we aimed select most stable reference genes from seven housekeeping (Actin, B2M, GAPDH, RPL13, EF1A, 18S UBCE) applied two of (GPS GPHK) under both normal physiological conditions stimulated lipopolysaccharide (LPS) or polyinosinic:polycytidylic acid (Poly I:C) relying quantitative real-time PCR (qRT-PCR). Additionally, raw Ct value resulting qRT-PCR was analyzed by geNorm, NormFinder BestKeeper algorithm, results indicated that expression all candidate exhibited some discrepancy different experimental types. As non-stimulated group, RPL13 were appropriate GPS GPHK cells, respectively. Nevertheless, suitable stimulation LPS, 18S, respectively, whereas after being with Poly I:C, UBCE EF1A recommended as optimal candidates To sure reliability selected genes, immune-related (ISG15, BCL2, IRF1 IRF7) chosen target normalize. will provide a direction various screen set stage application these relevant research areas.
